Chemical and Molecular Data

What is Cardiogen 20MG?

Cardiogen (H-Ala-Glu-Asp-Arg-OH, also called Ala-Glu-Asp-Arg, AEDR, the AEDR tetrapeptide and the Cardiogen bioregulator) is a synthetic tetrapeptide built from four L-amino acids: L-Alanine, L-Glutamic acid, L-Aspartic acid and L-Arginine. It belongs to the family of short peptide bioregulators, peptides two to seven residues long, and it is listed in PubChem CID 11583989 as C18H31N7O9 with a molecular weight of 489.5 g/mol.
Cardiogen is the Pinealon sequence, Glu-Asp-Arg, with one L-Alanine added at the front, and it shares its first three residues, Ala-Glu-Asp, with Cartalax and Bronchogen. In a 2023 docking survey of 26 short peptides it ranked 3rd of 26 at both the LAT1 amino acid transporter (ICM-Score −35.35) and the PEPT1 peptide transporter (−27.50), and fluorescence assays show it binds histone proteins H1, H2B, H3 and H4.

What are the Mechanisms of Action?

The biochemical targets and binding logic of Cardiogen in laboratory research include:

  • Predicted binding at the LAT1 amino acid transporter: In a docking survey of 26 ultrashort peptides, AEDR ranked 3rd at the LAT1 binding site with an ICM-Score of −35.35 (PDB 7DSN). That score is more favorable than every reference LAT1 ligand in the same study, including the potent inhibitor JPH203 (−31.50), L-Leucine (−23.96) and BCH (−17.37) (Khavinson et al., 2023).
  • Predicted binding at the PEPT1 peptide transporter: At the PEPT1 di- and tri-peptide transporter, AEDR also ranked 3rd of 26 (−27.50, PDB 7PN1), behind only Pinealon’s EDR (−37.59) and Prostamax’s KEDP (−33.42). That is level with the strongest reference PEPT1 compound tested (−27.74) and well ahead of the other three (−12.75, −11.64 and −7.90) (PMC10046148).
  • LAT2 docking: At the related LAT2 transporter, AEDR ranked 7th of 26 (−27.54, PDB 7CMI), close to L-Leucine (−27.78) and behind JPH203 (−34.69). The authors list AEDR among the most effective LAT2 ligands in the set (PMC10046148).
  • The C-terminal Arginine matters: Within the same survey, the Ala-Glu-Asp peptides differ only in what comes after Asp. At LAT1, AEDR scored −35.35, AEDG −32.93, AED alone −28.14, AEDL −25.11 and AEDP −20.95; at PEPT1, AEDR again led the group (−27.50 versus −25.35, −18.95, −18.00 and −12.65). The authors note that the best transporter ligands carry a neutral or positively charged C-terminal residue, and Arginine is the positively charged case (PMC10046148).
  • One Alanine added to Pinealon: Removing Cardiogen’s N-terminal Alanine gives EDR, the Pinealon sequence and the top-scoring peptide at LAT1 (−45.32) and PEPT1 (−37.59). Adding that Alanine shifted both scores by about ten units in the less favorable direction (PMC10046148). A reasonable inference is that the extra residue changes how the charged Glu-Asp-Arg core sits in each pocket, though the paper does not test that explanation directly.
  • Histone binding: In fluorescence quenching measurements, AEDR bound FITC-labeled histones H1, H2B, H3 and H4. The binding was traced to the N-terminal histone regions, depended on the histone and the peptide sequence, and was described as site specific (Fedoreyeva et al., 2013).
  • DNA and endonuclease interactions: A 2021 literature summary lists AEDR among the short peptides that either inhibit or stimulate hydrolysis of λ phage DNA by the WEN1 and WEN2 endonucleases, depending on the methylation state of the DNA (Khavinson et al., 2021).
  • Mixed-charge chemistry: Cardiogen carries two basic groups (the N-terminal amine and the Arginine guanidino group) and three carboxylic acids (Glu, Asp and the C-terminus). That is why PubChem counts 9 hydrogen-bond donors and 11 acceptors for C18H31N7O9, with a polar surface area of 290 Å2 and a computed XLogP3 of −6.3.
